Step: Unstained smear

Start with an unstained smear: both Gram-positive and Gram-negative cells are transparent and difficult to see. Apply crystal violet to flood the slide with purple dye.

What this sim shows

A mixed smear with Gram-positive (thick peptidoglycan) and Gram-negative (thin peptidoglycan + outer membrane) bacteria.

Step-by-step logic (under the microscope)

Unstained smear: both cell types are nearly invisible. This highlights why Koch and others developed staining — to turn transparent cells into high-contrast shapes.

Quick activities

Run through all four steps in order. Ask students to call out which cells stay purple vs. turn pink.
Reset and stop after decolorizer. Discuss which group has lost crystal violet and why.
Connect the final colors (purple vs. pink) back to cell wall structure and diagnostic Gram stains.
